In this paper , the relationship between cyanide - resistant respiration and expression of alternative oxidase ( aox ) using polyantibodies prepared by synthetic polypeptide in mung bean seedling , as well as the effects of vernalization on cyanide - resistant respiration and expression of aox in winter wheat were studied respectively . in the first part , twelve peptides , including eight conservative amino acid residues in the amino acid sequence of hydrophilic s helix of aox . were synthesized by solid - phase method 第一部分,我們按照交替氧化酶( alternativeoxidase , aox )位于線粒體內膜外側親水區s螺旋的氨基酸序列,用固相法合成由12個氨基酸殘基組成的多肽,將此多肽與-牛胰凝乳蛋白酶原a相連,用作半抗原,免疫家兔制備人工合成12肽的抗體。

The valt and the p valt of cyanide - resistant respiration were induced dramatically as well as the total respiration in early stage of vernalization treatment and deceased subsequently . the western blotting results also showed the parallel changes . during the whole treament time of 0 - 30d , although the cyanide - resistant respiration was increased , its opearation was lower than the cytochrome pathway which also made the main contribution to total respiration of winter wheat 結果表明:冬小麥胚芽在春化處理初期,伴隨著總呼吸速率的上升,抗氰呼吸容量( valt )和實際運行活性( pvalt )也快速升高,但在春化后期兩者均呈下降趨勢,與westernblotting結果十分吻合;在整個春化處理期間,抗氰呼吸活性雖有所上升,但相對于細胞色素主路來說,其運行還是處在一個較低的水平,而細胞色素主路則仍然占據了優勢。

In the second part , the winter wheat embroy under vernalization treatment was used to study the effects of non - stress - vernalization low temperature treatment on the development and operation of cyanide - resistant respiration and expression of aox 第二部分,我們以冬小麥為材料,研究了不同時期非脅迫春化摘要低溫處理條件下冬小麥胚芽抗氰呼吸發生、運行和交替氧化酶表達的變化。

We concluded that the increasement of cyanide - resistant respiration in the early stage of vernalization might be related to the adaptation of low temperature when winter wheat lives through the winter 據此我們認為春化處理對抗氰呼吸的誘導只表現在初期,并與冬性植物越冬初期的冷適應有關。
