This paper focused on two aspects : the physiological effects of cold stress on the euonymus japonicus “ zhuzi “ ; the isolation of cold - induced cdna gene . the results obtained were as follows : first , the chlorophyll ( chi ) and carotenoid ( car ) contents in euonymus japonicus “ zhuzi “ seedlings decreased greatly as the cold stress proceeded , accompanied by the sharply increasing contents of mda and pmp . it shows that chi and car degradation under cold stress was correlated with the damage of the plasma membrane 主要研究結果如下: ( 1 ) ‘粗枝’大葉黃楊在低溫脅迫下,隨脅迫時間的延長,葉片中脂質過氧化物的終產物( mda )含量明顯增加,細胞質膜相對透性( pmp )明顯增大;葉綠素( chl ) 、類胡蘿卜素( car )含量降低,且其含量的降低與mda含量增加及pmp升高間呈顯著負相關,說明低溫脅迫下‘粗枝’大葉黃楊幼苗chl和car的降低與生物膜的損傷有關。 |

Furthermore , suppression subtractive hybridization ( ssh ) was employed for the isolation of cdna fragments for euonymus japonicus “ zhuzi “ differentially expressed genes , and forward suppression subtractive cdna library of cold - regulated genes was constructed . the seedlings of euonymus japonicus “ zhuzi “ treated with low temperature were as tester and untreated seedlings as driver . subtractive cdna library was differentially screened through cdna macroarray , six hundreds and four cdna clones were identified as cold specifically induced or highly expressed ( 5 )應用抑制差減雜交( suppressionsubtractivehybridization , ssh )方法,構建冷誘導表達的正向抑制差減cdna文庫,低溫處理的幼苗為tester ,常溫處理為driver ,通過cdna微陣列差異篩選cdna文庫,得到604個低溫誘導或表達增強的候選克隆,對其中的84克隆進行dna測序,去除冗余的cdna ,在genbank中進行核酸和蛋白質同源性的比較和功能分析,共有36個單一序列,其中12個cdna在genbank數據庫沒有同源的序列。 |
